In the beginning when Mizu grabs the knife they smirk a little. It is because they notice their own signature on the knife. It adds another dimension to their words. This series is absolutely full of little details like that.

I've seen a lot of good discussions in the fandom identifying them as nonbinary, a trans man, or a cis woman; and the show doesn't really discount any of them (I lean towards nonbinary).

(And you may have already figured it out too, but I figured the song playing during Mizu's training montage (Tomoyasu Hotei's "Battle Without Honor or Humanity") was both appropriate to Mizu's moral code and a shout-out to Beatrix Kiddo in Kill Bill.)
