After giving myself a routine covid test I decided to explore the test strip.

I deliberately kept it simple and avoided the word "antibodies" which is the basis of the keys that attach the dye to the virus and then catch it as it passes the test line.   I felt that the word might trigger the flat earthers.

The Control line appearing is normal with just the buffer solution. It's designed to show enough of the sample has moved up the strip for the actual test to be accurate. Low pH seems to affect the false positive rate. https://www.sciencedirect.com/science/article/pii/S1201971221006548

The C line showing with just the buffer may be the origin of some of the false negative reports. I’ve heard (from a doctor) of the same thing happening with the PCR test when the swab isn’t done correctly.

The first couple of tests I did I didn't put the swab far enough back in the throat and the control line didn't appear, so the buffer solution alone doesn't appear to be enough (at least it wasn't for me).

I use the nose only veriety, jamming the swab up your nose until it almost comes out my ear is better than gagging on the throat veriety. Very interesting Clive and something else I don't have to wonder about. I can be the smartarse down the pub that knows how they work. Nice one Clive, thanks.

On a side note to this, I found out that those test strips are actually made of nitrocellulose aka gun cotton! I did try setting a dry one on fire, but it seems all the other chemicals on it stop it from getting "exciting"

There's at least one other version of the kits being distributed by the NHS. Same basic process but that one doesn't ask you to swab the throat, just the nose. No gagging, but the swab in that one is bigger and rougher, so it feels like your nose is being probed by aliens.

Fascinating, I kinda deduced this was how it worked but didn't know for certain.

Keep testing Clive. I had symptoms of a bad cold for 3 days, tested on day 1 with a -ve result. Did a routine test on the 3rd day as I was going to fly to Inverness and bang... The T line showed up as soon as the buffer/sample fluid hit it. Hope you feel better soon.

I was disappointed to find that the Control line doesn't actually test for the presence of DNA or anything, so simply using a couple of drops of neat buffer solution gives a result that looks negative. It makes you wonder how many infected people have failed to take a proper sample, but have then been greeted with a negative result.

The Binax test I used available in the States is a little different but essentially the same principle. This one had you just swab your nostrils. You placed the carrier solution directly on this cardboard card where there was a well and swirled the swab around a bunch. Then you folded the top of the card over onto the bottom part with the well leaving the swab in, it had a clever little cutout for it to rest horizontally. The top has the paper strip and then it soaks through. Takes 15 minutes. I was positive despite being vaccinated and the line was bright red at 7 minutes. Luckily it was a very light case and I now have a boost of immunity because of it. I work with the public so it isn't exactly surprising I contracted it.
