Anakin/Vader found out about Luke from Palpatine in Empire Strikes Back. Palpatine says that Luke is Anakin's son and Anakin/Vader is like 'its impossible' or w/e and Palpatine says 'you know it to be true'. Obi-Wan told Luke in Return of the Jedi that he has a twin sister, I'm pretty sure and through the Force, he was able to instantly know it was Leia. Also, Return of the Jedi is when Vader/Anakin found out that Luke has a sister, but it's not clear he knew her identity. It just seemed like he could read Luke's thoughts just enough to know that there's a sister somewhere he's protecting. No one knew about Leia being Anakin/Vader's daughter in A New Hope, but they did say that she was resistant to the mind probe, which is just kinda foreshadowing that she has a connection to the force.
