From a few episodes ago, when they were first exploring the tunnel after freezing Sloth outside, Ed realized the tunnel went around the entire nation. On the map they were using they realized that the places that overlapped with the tunnels line were all places that major conflicts had happened, resulting in a lot of lives lost and blood spilled. Similarly a line was drawn connecting those places and those secondary lines had similar locations, the only place not having an incident like that was Briggs. So a crest of blood seems to at the very least be a marker for the specific/important points of the Nationwide Transmutation Circle. I'm trying to be a bit vague because I don't remember if they show has explained it yet or not.
