Once Luffy knows that Sanji is aware of pudding, he knows that Sanji has fully agency to make his own decisions. Luffy knows that Sanji will comeback, but is only afraid he maybe trick if is lied to by pudding.

I think most people know that in the real world, a third eye is a reference to enlightenment and spiritual connection, but also (I think) in taoism the third eye is connected with memories. Just fun references and inspiration Oda takes from all kinds of things.

It's more like he can sense or feel what the message is trying to convey. The only things that we've learned about the three-eyed clan are that they can awaken an ability similar to the voice of all things which would allow them to decipher poneglyphs, and we also know they are one of the few races left in the world that are considered rare. Based on their rarity, you can speculate that the World Government probably hunted them down over the last 800 or so years, whether their abilities were real or not.

Also she was asking Pudding if she had awakened the power of her third eye, which is separate from her Memo-Memo Fruit power, to which she complained that she was only a half-blood and might never get that power. I don't know if Big Mom is unaware of Pudding's ability to alter memories (useful devil fruits are so rare that I doubt that she would miss Pudding getting something like that), but Pudding's "innocent act" was more for the chesskeepers and the castle guards. Remember, Pudding told Reiju, "Only my family is aware of my true nature." That's why she asked her mom to speak privately in her room, so they could talk more openly.
