の (no in romaji) is a particle marking the genitive, which can sometimes be translated as "his" though here it simply means "of". その (sono) is the bit that means "that"

Yes, the direct translation would be "that". However, I've mostly seen it translated into that as it seems its meant to be referring to both Dio and Johnathan, thus why its often translated that way as the rough translation.

"No" refers to a man? I always thought that "sono chi no sadame" meant "destiny of the/that blood" and "sono chi no kioku" meant "memory of the/that blood"
Then again, I'm not japanese.

The new opening is called Sono Chi No Kioku, which roughly translates to "Memory of his blood" as compared to "Sono Chi No Sadame" meaning "Destiny of his blood". Which is fitting as this is the big return Dio, who's conflict with Johnathan started all of this.
