For the last question (elaborate on the inciting situation):
The boys (i.e. the turtles) are cornered, and April has to save them. She'd previously gotten ahold of new experimental mutagen that Krang was going to use to make an army of dino-soldiers. With no other option she doses herself hoping it would give her something to help save her friends. Could add a throwaway line about how Krang was going to discard this formula because while it left the subject intelligent they also still had their free will, or something like that.
